pseudomonas aeruginosa biofilms.. i ve onl basic knowledge abt wat biofilm is..
so pls help me in carryin out this proj.. i wanted to knw as how p.aeruginosa
can be grown as a biofilm in laboratory condition.. is it possible 2 get a
large sample by growin d culture under batch conditions instead of growin in
continuous tubing reactor as we dont hav that set up facilities in our lab.. if
so can u suggest me d expt methods fr biofilm formation & screening.. i
wanted to work on biofilm associated proteins, its structural
characterisation.. m concentrating in proteomics to study by 2D- PAGE.. pls
reply me ur

Hello Mr. Wright,

I'll begin by apologizing if I have addressed you incorrectly, I came across your name in a rather dated (1997) correspondence with George O'toole when searching for a good way of enumerating the viable cells in a biofilm. The method you describe to Mr. O'toole (employing sonication of cells followeed by dilution) sounds perfect my intended application;enumerating the E. coli cells from a biofilm in a microplate, and was hoping that you may have on hand a reference for the protocol you described. Here is a link to the description you provided http://www.bio.net/bionet/mm/biofilms/1997-April/000106.html.

I am looking for an standard method to measure Bulk Modulus of Biofilm. I did not find any method in the previous literature regarding this issue..

Would you please suugest whatever you think would be useful for this type of compression test (in hydrostatic stress condition)?

The initial test results suggests to me that biofilm can be treated as a nearly incompressible material (Poisson's ratio of 0.49 N/m^2), this is assumed by some other people (0.5) and others assumed 0.3 which suggest that biofiulm is compressible...I am not clear what value I should use for simulation

font-family:Arial'>Dear all,

<span style='font-size:10.0pt;

I am a mechanical engineer who is measuring biofilm
mechanical properties. I am currently doing compression tests on mixed biofilm.
I have two questions:

Biofilm used in the test is a
few weeks old, do I expect to see big difference in results when I do it
     on very fresh biofilm?

I had a 100N load cell and I
know sensitivity is low, does it affect results in unacceptable way?

Biofilm is placed in a glass
container and I have a surface tension problem and biofilm sticks to the
glass wall, any recommendation to reduce adhesion? I used PTFE lubricant
     but didn’t work well.

<span style='font-size:10.0pt;

Chemical Engineering and Applied Chemistry, University of Toronto, Toronto, ON M5S3E5. This landmark paper is the first example of a careful and thorough use of Phenotype MicroArray data to challenge and improve a whole-cell bioinformatics model of Bacilllus subtilis. Network gap analysis was used to identify 48 essential reactions missing from the genome annotation. Detailed growth rate analysis revealed incorrect modeling outcomes which were subsequently improved by adding 75 reactions. Simulation of growth phenotypes of knock-out strains were verified in 720 of 766 cases. Overall, the integrated analysis of substrate utilization and gene essentiality data led to identification of 80 required enzymes and improved the genome annotation for Bacillus subtilis.

Department of Microbiology, Cornell University, Ithaca, NY 14853-8101, USA. The biological role of extracytoplasmic function sigma factors in Bacillus subtilis was examined in this paper. Seven genes encode these sigma factors, so the authors constructed strains with multiple gene knockouts and then used Phenotype MicroArray technology to test the phenotypes of their strains. A triple mutant in sigM, sigW, and sigX exhibited greatly increased sensitivity to detergents, polymyxin B, B-lactams, and D-cycloserine.

Included in the tests are assays of basic cellular nutritional pathways for C, N, P, and S

metabolism, osmotic and pH and sensitivity, and sensitivity to chemical

The most common application is to assess the

phenotypic effects of mutations. A change in genotype of a cell should lead to one or more

changes in phenotype. PMs allow testing of knock-out or knock-in mutants to help discern the

biological changes that occur consequent to genetic changes.

Abstract submission deadline: 17 September 2007

Early registration deadline: 22 August 2007

Final registration deadline: 21 November 2007

Full paper submission for publication: On-site (28 November 1 December)

CONFERENCE PUBLICATIONS

Proceedings Book

A multi-volume book will be published including full papers of works (oral, posters and virtual posters) presented at the Conference. The book will be published by an international publisher in order to give it a broad international distribution and to serve as a good reference to consult the state of the art of the hottest topics in Modern Microbiology. The paper length will be 5 printed pages. Submission of full papers is optional.
Abstract Book
A separate Abstracts book will be published with abstracts of works presented at the Conference. It will be distributed among full rate participants at the beginning of the Conference, and dispatched to virtual participants after it.
International Journals special issues
Agreements are being negotiated with several international journals, in order to produce special issues based on very good papers presented at the Conference. Manuscripts must be delivered during the Conference, according to the instructions that we will give in due course for each journal. All papers will be strictly reviewed and treated as regular papers. Our goal is to produce high quality and high impact special issues based on some of the best conference papers. Papers submitted for a journal special issue and rejected (for scientific/editorial/space reasons), can be considered for inclusion in the Proceedings book. Please refer to the conference website for details.
If you would like to know more information about the Proceedings Book and the International Journals special issues published after the 1st edition of this conference, BioMicroWorld2005 held during 15-18 March 2005, click here

VIRTUAL PARTICIPATION

In addition to the "traditional" oral contributions and posters presentations, a Virtual Participation modality has been established for those researchers unable to attend the Conference personally. Please consult the conference website:
http://www.formatex.org/biomicroworld2007/virtual/

COLLABORATION PROJECTS: SEARCH FOR PARTNERS

BioMicroWorld2007 will also serve as a platform for partners search for trans-national collaboration projects, like for example the EU Seventh Framework Program (Networks of Excellence, Integrated Projects and Specific Targeted Research or Innovation Project). All ideas/proposals for collaboration will be included in a booklet which will be distributed at the beginning of the Conference and/or posted at the conference website. If you are interested in taking part, please send us the corresponding form available at the website.

RESEARCH GROUPS

Send details of your group activities (research topics, members, projects, publications...). The booklet with all participating groups will be distributed at the Conference and/or posted at the conference website.

For any questions or suggestions, please do not hesitate to contact us at biomicro2007@...
We would also appreciate if could disseminate this Call for Papers through your Department or Institution.
We hope to meet you at this large interdisciplinary international meeting!

hello everyone!

May be I am asking a very simple and naive question, but I have a
doubt regarding the terms we use for checking antibiotic sensitivity
of biofilms. For checking antibiotic sensitivity of planktonic cells,
we use the terms MIC (minimum inhibitory concentration) and MBC
(minimum bactericidal concentration). Can we apply the same terms for
biofilms also? Or do we use the terms MBPC (minimum biofilm preventive
concentration) and MBEC (minimum biofilm eradication concentration) as
markers for antibiotic sensitivity of biofilms?
> While in theory any stain that would stain biological components of
> biofilms (and not the underlying polystyrene substratum) could be
> used, crystal violet absorbs much more light than safranin and so

While in theory any stain that would stain biological components of
biofilms (and not the underlying polystyrene substratum) could be
used, crystal violet absorbs much more light than safranin and so
would provide higher sensitivity for this assay.

Good luck with your work,
Surface energy is a measure of disruption of chemical bonds that occurs
when a surface is created. Most of the hydrophobic surfaces are known
to show low surface energy values. It is also well known that
hydrophobicity has a role in cell attachment to the surface and biofilm
formation. As the biofilm progresses the adhesion is higher and
chemical bonding is more hence lower the surface energy.

Few studies were carried out on adhesion of biological systems such as
proteins, microbes, algae and invertebrates. They all show minimal
long-term adhesion associated with surfaces having low energy surfaces.
However, all surfaces rapidly become modified on immersion in natural
waters through the adsorption of conditioning films, which may
influence subsequent adhesive events associated with the permanent
attachment of organisms. The results show that surface energy can
elicit different responses in different organisms. For most organisms,
minimal adhesion is associated with low surface energy.

According to thermodynamics, matter seeks to be in a low-energy state,
and bonding reduces chemical energy. Young's Modulus says, if the
surface is hydrophobic then the contact angle of a drop of water will
be larger. Hydrophilicity is indicated by smaller contact angles and
higher surface energy. Water has high surface energy by nature; it's
polar and forms hydrogen bonds. Hydrophilicity is generally indicated
by smaller contact angles and higher surface energy. Water has high
surface energy by nature; it's polar and forms hydrogen bonds.
> > Ref:http://www.bio.net/bionet/mm/biofilms/1998-September/000364.html
>
> > I am having trouble understanding the concept of surface energy (SE).
> > SE seems to mean the same thing as bonding potentiel.
> > What confuses me is that while hydrophobicity increases as SE
> > decreases, adhesion seems to increases. Why does adhesion increase
> > when the bonding potentiel (SE)decreases. I thought that the trend
> > should be the reverse. Moreover, I think that it contradicts the
> > hydrophobicity theory: adhesion of water decreases as the SE decreases.
>
> > Please make it clear for me; maybe I am missing out on something.
>
> > Best regards,
>
> > Alfred
