Help in purifying unknown Drosophila membrane protein.

Dear Dr. Ericsson,

I am a graduate student at the dept of Genetics & Neurobiology,
Wuerzburg Biocenter. I am working under Prof. Erich Buchner. I am trying
to purify and identify an unknown protein from Drosophila (10 kDa in
size) against which we have a monoclonal antibody, which is a one among
the Wuerzburg Hybridoma Library antibodies. It has not been possible to
pull down my protein by usual IP protocols, and then I found that when I
homogenize heads in a detergent free buffer and spin homogenate at
100000xg my protein remains in the membrane pellet. Cloud point
precipitation of the membrane pellet dissolved in 2% TritonX114, showed
that my protein remains exclusively in the detergent phase. Hence we
believe it to be a membrane protein. As a result 2DE attempts (using 7cm
stip Zoom IEF-PAGE from Invitrogen) also failed. In the mean time, we
found interesting staining pattern of the antibody in specific layers of
the medulla, lobula and lobula plate, whole ganglion (SOG & thoraic). It
is also present in the posterior most cell of the oogonium (egg cell)
and so gives strong western blot signal with embryos both within female
abdomens and after being laid as eggs. The protein is also detected in
these parts by immunostaining of cryosections. And some life stages like
larvae show a second higher (just above 10kDa) band in westerns. We are
very eager to study this protein which has such a small size, but
widespread but specific distribution and so early in life cycle.

So in this regard I would like to request you to help us in purifying
this protein (2DE or chromatography followed by MS or whatever you feel
is suitable). We can send our monoclonal antibody or if needed I can try
to come over, whichever is most convenient to you.

Querry regarding silver staining for glycoproteins

Agarose gel electrophoresis on glycoproteins

Hello everybody,

I am using agarose to study the purity of mucin glycoproteins. When I did the electrophoresis, no bands were detected with Coomassie blue even the marker (6,500- 205,000 Da), neither with Periodic acid Schiff staining. Could anybody who did the agarose gel electrophoresis on mucins help me? There was a kind of agarose called Metaphor XR that showed to be effective in separating glycoproteins but the company (FMC Corp) does not sell the product anymore. Please let me know if there is any substitute?

Dear Bin-I rarely work with the a2, 6-sialyltransferase anymore and I
don't know where you might be able to buy it. Why don't you try the
Consortium for Functional Glycomics.

I know they make glycans, glycan binding proteins, and antibodies. Even
if they don't have purified enzymes for sale, someone involved may have
in the course of their work purified the sialyltransferase and will

I’ll appreciate receiving information regarding the sialidase inhibitor 2,3 dehydro N- acetyl neuraminic acid:

1. Is it being used in industry as a supplement to culture medium in order to avoid cleavage of sialic acid residues of recombinant glycoproteins expressed in CHO cells?

uronic acid/total sugar ratio

Hi,

I work with small amounts of plant tissues and want to normalize amounts of uronic acid in my samples per something other then weight. I measure total sugar with phenol/sulfuric acid assay. Can I normalize the amount of uronic acid per nm total sugar?

In Arabidopsis there is a pectate lyase-like gene. The wild type plants with this gene are susceptible to powdery mildew pathogen, but the mutant plants without this gene are resistant to this pathogen.

Would you please help me to answer the following questions?

What are the possible function of this gene in plant disease resistance?
Please list as many possible functions as possible.
Please suggest as many resistance mechanisms as possible.

Postdoctoral position is available to study the function of two gene families involved in the synthesis of nucleotide-sugars in plants. The project involves cellular and molecular biology methodology in addition to biochemistry and analysis wall mutants. Candidates must hold a Ph.D. in molecular biology or biochemistry with an excellent publication record and strong credentials in modern molecular biology/genetics and enzymology/biochemistry. Experience with biochemical techniques such as heterologous protein expression, protein purification, enzymatic assays, antibody production, and experience with molecular and cellular techniques such as cloning, construction of fusion proteins (GFP), and immuno-EM protein localization is desirable. Salary commensurate with experience.
