We have a Biolam microscope still in the original case as yet unpacked. It was purchased in the 80s and passed along to us thru an estate. It was never

opened as the owner, Dr. Robert C. Beck, was waiting for an Instruction Manual in English. A plate on the microscope says: "nomo, Biolam, 6monam"

Do you know where we can get a manual in English? I would appreciate any help you can give us. Thank you, Carole

Workshop announcement - June 3rd-5th 2009

Dear All,

Cryptonet (www.cryptonet.ie) is pleased announce a three day practical workshop on molecular techniques and risk assessment focused on surface water contamination by Cryptosporidium spp. from agricultural sources. All are welcome to join at University College Dublin, Belfield, Dublin, Ireland from Wednesday 3^rd to Friday 5^th June 2009. The workshop will be hosted by the UCD School of Agriculture, Food Science and Veterinary Medicine, and The Central Veterinary Research Laboratory (DAFF). Participants can choose to join just the one day symposium on molecular methods and risk assessment and, if they wish one of two follow on workshops detailed below.

Contamination of surface water by Cryptosporidium spp.
A workshop on molecular biological techniques and risk assessment

A one day symposium bringing together methodologies in molecular identification of Cryptosporidium spp. and microbial risk assessment will be held on Wednesday June 3^rd 2009.  A combination of international and local speakers will present on key research issues relating to using molecular tools and risk assessment for Cryptosporidium spp. in agricultural catchments.  Attendance at the symposium is welcome without participation in the concurrent practical workshops to be held on June 4^th-5^th 2009.

Workshop 1: Molecular Techniques

A two-day practical workshop on molecular biological techniques: towards the development of standard protocols for the detection and quantification of Cryptosporidium oocysts in various environmental matrices. This session will cover the DNA extraction from complex environmental matrices (soil, slurry and contaminated water) and the detection of Cryptosporidium spp. using real time qPCR techniques. Special attention will be dedicated to statistical analysis of real time PCR data.

Workshop 2: Microbial Risk Assessment

A one-day workshop on risk assessment techniques applied to the study of non point source agricultural pollution. Using a participatory-based approach to teaching and learning, attendees will have the opportunity to use software tools and contribute to in-depth discussions, share experiences, and exchange resources in a variety of thematic topics such as Microbial and Ecological Risk Assessment, Risk Based Site Evaluation, Cryptosporidium Epidemiology. Participants will develop a sound understanding in developing conceptual risk assessment models and semi-quantitative methods in describing microbiological risk.

Ciliate morphometrics should account for cycles

Morphometrics fails to identify closely related ciliates that have the
same appearance, mostly because they vary in size through the clonal
cycle. Humans recognize shape, whereas computer programs like
Fisher-Mahalanobis Discriminant Analysis commonly are prone to recognize
by size. Has-or-has-not produces rather few characters.

The morphogenesis of ciliates uses random distribution statistics for
the taxonomy of ciliates, although the cell is cyclic and determined.
The arithmetic mean (AM), the standard deviation (SD) and the mode (M)
are determined, and so are genetic factors including clonal cycle
effects. Some cell regulation has long been developed in Paramecium by
James D. Berger and his associates.

It is now suggested that the mode of distance units with 10 %
percentiles is the most informative and logically applicible descripter.
Trigonometric variables should be used in morphometrics.

A new NSF-funded postdoctoral position at the Bigelow Laboratory for Marine
Sciences offers an enthusiastic and highly motivated scientist the exciting
opportunity to work with us in a cutting edge, globally important and
technologically advanced research area of marine genomics. Bigelow is an
international center of excellence for microbial oceanography and holds the
world's largest collection of phytoplankton cultures. Bigelow's research
portfolio places particular emphasis on the use of molecular and
post-genomic tools to understand the role of marine viruses in microbially
driven biogeochemical processes.
A Marine Molecular Biologist / Virologist is sought to develop and utilize a
virus oligo-microarray from a recently sequenced marine algal virus.
Transcription profiles from the virus and host genomes will be generated
during in vitro and in situ infection experiments to look at the role of
nutrient availability on infection. A single-cell genomics approach will
also be developed to look at diversity of viruses from single infected cells
from environmental samples. The candidate will be expected to participate
in shore-based fieldwork in Norway (1-month). There may also be research
cruise opportunities in the Gulf of Maine and the Atlantic Ocean therefore
you must be willing to work flexible hours at sea, if required.
We are looking for a research scientist interested in the role of viruses in
marine ecosystems. Applicants must have a PhD or equivalent in either:
virology, microbiology, molecular ecology and/or post-genomic technologies.
Working experience of bioinformatics software will be a definite advantage.

Hello Dr. Goodkov

Tonight while surfing I happened across your March 1999 message remarking that you studied protozoan behavior as an undergraduate and graduate. For my Master's degree in psychology I conducted experiments to determine whether paramecium aurelia's behavior could be conditioned to time intervals using a classical conditioning paradigm, with a weak electric current as the UCS. 3/5 experiments were significant---after training, the beasties clustered around the area of the + pole at intervals approximating those framing introduction of the UCS.

It is, as you say, a fascinating field. Late nights all alone with a test tube...I will follow the references you gave, thank you.

Im working with the pathogen Cryptosporidum parvum, the protozoan
found in runoff waters from agricultural practises. They are primarily
found in the feces of the calves and regarding the culturing methods
you may refer any of the research papers studying cryptosporidium
transport.

Ciliate strains, antibodies available --

To all protist colleagues: Due to my impending retirement from active
research on my pet ciliate, Euplotes, I am disbursing my stocks of
several Euplotes species, and one Oxytrichid. The attachment lists
those strains available, and their origins and preferred (lab) foods.
All are freshwater, although one (E. woodruffi) was adapted from a
local brackish water collection.

If anyone is interested in receiving any of these strains, please
contact me (best first by phone) and I can ship them to you. I will be
moving across country this spring, and do not intend to take any cells
with me.

Soon I will also be dispensing some of my stocks of antibodies against
Euplotes cytoskeletal proteins that I have made. Monoclonals and
polyclonals against PLATEIN proteins, and polyclonals against a basal
body scaffold protein that I have dubbed CAGEIN (Mss in preparation).

Seeking research on entomopathogens

I'm currently a graduate student at Portland State University working on a
literary nonfiction thesis about butterflies. (Science writing for a
general audience.)

Throughout my thesis one of the themes I'm exploring is what butterflies
and humans have in common. In the chapter I'm currently researching, I'm
focusing on entomopathology, particularly things that attack Lepidopteran
larvae in diapause (in the field as well as in an artifical environment
such as a Petri dish).

Macronuclear regeneration

Discovered in 1940 in Paramecium aurelia by Sonneborn, macronuclear
regeneration (MR) in ciliates is now well established. Senescence,
immaturity (Haga & Hiwatashi, 1981) and rejuvenation have entered the
panorama of conjugation and autogamy even since Maupas in 1889. See
Smith-Sonnenborn (1979) on aging.

A week ago, I made the mistake of writing that MR was not convincing,
corrected now after reading Kloetzel on Euplotes aediculatus and Kimura
& Mikami on Paramecium caudatum. If the initiating macronucleus (MA) is
damaged or missing,, a MA fragment of the old MA can replace it as a
form of life insurance. Kloetzel used a UV microbeam and Kimura & Mikami
used microsurgery to inactivate the anlagen and induce MR from fragments.

Ciliates are multinucleate cells that contain macronuclei and
micronuclei. The nuclear contents often change after meiosis. Crossing
over, various segregations and hybridizations often involving
allopolyploidy are still poorly understood. Exconjugant cells have
maternal MA fragments, newly initiated MA and micronuclei (MI). The
maternal macronucleus of P. caudatum is retained for long periods such
as four cell cycles in some species and can continue to influence
development.

However after a few cycles, the presence of the new MA anlage suppresses
the synthesis of DNA by the maternal fragments that then become pycnotic.

Then MA fragmentation might begin with fertilization or later followed
by MR followed by DNA inhibition in fragments and then their digestion.
See Kimura & Mikami (2003). Without a new MA, the old one is
regenerated, at least in both P. caudatum and Euplotes aediculatus.
Sending a genetic signal through the cytoplasm from an anlage to a
fragment is a miracle indeed. Is this one a somatic segregation ? Such a
signal was assumed to originate in the MI-controlled synkaryon as MIs
and MAs so segregate from it. That is the MIs from the synkaryon
segregate into MIs and MA anlagen. See also Cole & Siegel (1969) leading
to Chen on allopolyploidy in P. bursaria.

When ciliates say Make 4 MI, they might only need one. If the processes
of meiosis and allopolyploidy are accidentally mixed, one result can be
a new species in one jump though the common result leads to death.
Ciliates are, by the precautions they take, trying to hold on to the
status quo including of course their ploidy status. It is indicated by
the volume of the MI. Segregations in the sense of being only related to
crossingover in classic meiosis cannot explain all of the events during
the Maupasian rejuvenation allowed by conjugation--promoted by sexual
exchange.

As a matter of fact, this organism was a rotifer of the genus
Collotheca. Thanks to Dr. Nomdedeu. Thereafter, I was able to find a
lot of infos about that genus on the web (even very nice movies of
different species). I will try to determine the species with some
publications and papers on rotifers in Canada of which I found the
references.

Hello,

I know this posting is not appropriate here but protistologists are in a
better position than most biologists to give a sound advice about
organisms living in the microscopical world.

So, I recently found in a small freshwater pond something that is
probably a rotifer but one of a special kind I never saw before. If
somebody could give me a hint about its exact nature, that would be
greatly appreciated.
