Determination of particle/pfu ratio

Dear all,

I have OD 260 value and PFU/ml value of purified virus
sample. How then do I calculate particle/pfu ratio
using these values?

So, do you know how many virus particles corresponds to 1 OD unit?

If not, you will need to measure it. Electron microscopy? Or some
labeling.

If you know the size in base pairs of the virus, you can calculate the
molecular weight of the viral DNA (1 nucleotide PAIR= 660 daltons).
Viral DNA concentration (calculated from OD 260 value) can then be
converted to viral particles using Avogadro's number (1 Gram MW=
6.02e+23 viral particles).
