get of ura3 mutant

I tried to get an ura3 mutant of a yeast by EMS or MNNG mutagenesis and 5-FOA selection. But I could not get it. So I would like to get it by other method. Could you tell me other methods that create ura3 mutant of yeast.

Is it diploid or haploid? What about using homologous recombination of a PCR generated cassette?

I am not sure whether it is diploid or haploid, but I think that it may be diploid. Could you teach me in more detail the method using homologous recombination of a PCR generated cassette.
